DNA在哪裡？
•真核生物細胞中DNA會與蛋白質纏
繞形成染色質，在有絲分裂與減數分
裂前染色質會聚合形成染色體。

細胞膜、核膜：要打破
蛋白質：要分解
DNA：要溶解

打破細胞壁與膜
1. 取奇異果或洋蔥果汁
30ml
2. 加入1ml洗碗精攪拌5min

打破細胞膜與核膜
洗碗精中含有sodium lauryl
sulfate(十二烷基硫酸鈉)，可溶解磷
脂質。加入洗碗精可打破細胞膜與核
膜。

溶解DNA
1. 加入2.5ml的5M食鹽水，攪拌5min
2. 5M=50ml水+15g食鹽
3. 鈉離子可以中和DNA的負電荷。加
入食鹽水可以協助DNA的溶解

分解蛋白質
1. 加入2.5ml的5%嫩精，攪拌5min
2. 5%=50ml水+2.5g嫩精
3. DNA會與蛋白質纏繞，分解蛋白質
可得到較純的 DNA。嫩精含有分解
蛋白質的酵素

過濾與析出
1. 用紗布過濾混合液。
2. 取出過濾混合液5ml
3. 緩緩倒入5ml的95%冰酒精
4. DNA不會溶解於酒精中。

1. 酒精中白色絲狀物即析
出的DNA。
2. 將DNA取出放入針管
